摘      要:Drug addiction has become a major public health problem worldwide. Conventional treatments can ameliorate the Abstinence symptoms effectively,but have little to do with relapsing. Recently,ablating the brain regions related to drug addiction permanently through stereotaxic surgery has been proposed and used as a new therapeutic strategy in China. This bold attempt is highly controversial,and major concerns are about the efficacy and side effects of the surgery. In the present study,morphine-induced conditioned place preference (CPP) was established in rats as an animal model to imitate the addicts’ psychological dependence or reflect the craving for the drug of abuse. Direct current (2 mA,7 s) was delivered to ablate sub-regions of bilateral nucleus accumbens (NAc) via a stainless microelectrode,which was similar to the stereotaxic surgery used clinically. To investigate the effect of bilateral NAc core lesions on morphine-induced CPP and the possible side effects on animals’ general physiological function,four behavioral models,Morris water maze,sucrose-induced CPP,open field test and delay discounting task,were applied to assess the rats’ spatial learning ability,associative learning ability,exploratory behavior and impulsive control ability,respectively after the surgical lesions. Results showed that lesions of the NAc core,but not the shell blocked the maintenance of morphine CPP in rats. Although the rats’ spatial learning ability was not significantly affected,their associative learning ability,exploratory behavior and impulsive control ability were obviously impaired by the operations. Our findings indicate that lesion of NAc core blocked the craving for morphine,while impairs associative learning and impulsive control abilities in rats,and that suggests lesion of the NAc core or shell should not be routinely used as a therapeutic strategy for the treatment of opiate addiction.
